How Tofacitinib Works?
Tofacitinib inhibits Janus kinase (JAK) enzymes, JAKs activate signal transducers and activators of transcription (STATs), which regulate gene expression and intracellular activity. Inhibition of JAKs prevents cytokine- or growth factor-mediated gene expression and intracellular activity of immune cells, reduces circulating CD16/56+ natural killer cells, serum IgG, IgM, IgA, and C-reactive protein, and increases B cells.
Adult Dosing
Psoriatic arthritis
Immediate release: 5mg twice
Extended release: 11mg once
Rheumatoid arthritis
Immediate release: 5mg twice
Extended release: 11mg once
For hepatic and renal patients, it’s preferred to reduce the dose to once daily, and stop it in case of sever hepatic impairment.
